Determination of 5-S-cysteinyldopa in melanomas with a fluorimetric method.

چکیده
Differences in color of the skin and hair of animals have been the subject of much research. Such differences have been ascribed partly to differences in the relative amounts of dark eumelanins, and lighter pigments, phaeomelanins, in the pigmented tissues. The chemistry of eumelanins has been extensively discussed in the classic monograph of Nicolaus (1). The early work of Prota and Nicolaus suggested that phaeomelanins were derived from tyrosine and cysteine (2). All later studies lent support to their view. At the VIIth International Pigment Cell Conference in Seattle in 1969, Prota presented the review "Structure and biogenesis of phaeomelanins" (3), which summarizes the very important work in this field from the Department of Organic Chemistry in the University of Naples. In 1966 two catechols giving different fluorophores when oxidized were described in human melanomas (4-6). One of them proved to be dopa, while the other remained unidentified for many years. Recent work has shown that this second catechol is 5-S-cysteinyldopa (7-9), the compound considered to be the building-stone of phaeomelanins. This review concerns studies on 5-S-cysteinyldopa, which suggest that determination of this thiocatechol may be helpful in the definition of pigment metabolism in health and in disease.

Fluorimetric Methods for the Determination of Terazosin HCl in Drug Substance and Dosage Forms
Determination of terazosin hydrochloride dihydrate (TRZ) in drug substance and tablets was studied by fluorimetric techniques. The fluorimetric methods are based on: (1) measurement of the native fluorescence of the drug in water at 750 nm after excitation at 330 nm. (2) sensitizing the native fluorescence by formation a binary complex of drug with aqueous uranyl acetate (0.1% w/v) at the same ...
متن کاملExtraction-Fluorimetric Determination of Low Levels of Copper by 1,7- Diaza-15-Crown-5 and Rose Bengal
A simple and sensitive extraction-fluorimetric method for the determination of copper is reported. The 1,7-diaza-15-crown-5-copper-rose Bengal (DA15C5-Cu-RB) ternary complex is quantitatively extracted into chloroform, diluted with ammoniacal acetone and its fluorescence intensity is measured at 570 nm with excitation at 540 nm. A linear calibration graph was obtained over the copper concen...
متن کاملThe pigmented life of a redhead.
As a redhead I have had a personal interest in red hair, freckles and sunburns since childhood. An observation of a formaldehyde-induced fluorescence in human epidermal melanocytes initiated my scientific interest in these cells. Prota and Nicolaus demonstrated that oxidation products of cysteinyldopas are the main components of pheomelanin. Our identification of 5-S-cysteinyldopa as the source...
متن کاملOxidation of tyrosine residues in proteins by tyrosinase. Formation of protein-bonded 3,4-dihydroxyphenylalanine and 5-S-cysteinyl-3,4-dihydroxyphenylalanine.
A simple and rapid method was developed for the determination of 3,4-dihydroxyphenylalanine (dopa) and 5-S-cysteinyl-3,4-dihydroxyphenylalanine (5-S-cysteinyldopa) in proteins with the use of high-pressure liquid chromatography. With this method, it is demonstrated that mushroom tyrosinase can catalyse hydroxylation of tyrosine residues in proteins to dopa and subsequent oxidation to dopaquinon...
متن کاملExtraction-Fluorimetric Determination of Trace Amounts of Mercury by 1,10-Diaza-18-Crown-6 and Rose Bengal
A simple and sensitive extraction-fluorimetric method for the determination of mercury is reported. The method is based on the quantitative extractionof a ternary crown-mercury-rose Bengal complex into chloroform at pH 10 followed by fluorimetric measurements at 580 nm. A linear working range from 0.05 to 2.00 mg/mL is obtained. The relative standard deviation for 1.0 </em...
